ChIP-seq
Chromosomes are chemically treated in vivo to crosslink DNA and chromatin proteins. The cross linked material is fragmented and then bound by antibodies to specific chromatin marks or DNA-bound factors. DNA from this immunoprecipitated material can then be purified and subjected to Illumina sequencing. By mapping the resulting reads back to the original genome it is possible to infer the locations of the specific chromatin mark or DNA-bound factor.
In addition to sequencing, the GRH offers manual library prep using Illumina ChIP-seq reagents. The GRH can also offer fragmentation of cross-linked chromatin samples using the Covaris M220 ultrasonicator.
